The function of the primary catalytic residues Glu134 and Glu138 in retaining 1,3-1,4-beta-glucanase from Bacillus licheniformis was studied using a chemical rescue technique based on enzyme activation of inactive mutants by adding nucleophiles. Alanine replacements on both catalytic residues produced inactive E138A and E134A mutants. The activity may be reestablished by applying a highly reactive substrate with a good leaving group through the addition of a reactive nucleophile.
