ÚČINNOSŤ VYBRANÝCH RASTLINNÝCH SILÍC A EXTRAKTOV NA TVORBU EXTRACELULÁRNEJ FOSFOLIPÁZY CANDIDA ALBICANS. (Slovak)

Candida albicans is an opportunistic pathogen responsible for the development of candidiasis. Its pathogenicity is due to several factors that contribute to its resistance to conventional antifungals, thus worsening the prognosis of treatment. In our study we focused on the antifungal and antivirulent effect of plant extracts (PE) and essential oils (EO) from the family Lamiaceae (Mentha piperita L., Thymus vulgaris L., Origanum vulgare L., Salvia officinalis L.) on Candida albicans strains. Of the tested PEs, the best antifungal effect was observed in the extract from Origanum vulgare L. with an average minimum inhibitory concentration (MIC) of 4.9 mg.ml-1, therefore its effect on the production of extracellular phospholipase by C. albicans was further monitored. A better antifungal effect was observed with EOs at average MIC values of 0.4 mg.ml-1 (Origanum vulgare L., Mentha piperita L., Thymus vulgaris L.). Of the 23 isolates (92 %) producing EPL, 22 were tested for inhibition of EPL production by plant yields. After exposure of the cellsto the extract, 7 isolates(31.8 %) produced EPL intensively, 6 isolates (27.3 %) showed medium EPL production and 8 isolates (36.4 %) moderate EPL production. One isolate (4.5 %) did not show phospholipase activity. After one hour exposure of Origanum vulgare L. RS (0.8 mg.ml-1), only 1 isolate (4.5 %) continued to produce EPL intensively and up to 15 isolates (68.2 %) did not form phospholipase. Another 4 isolates (18.3 %) produced EPL moderately and one isolate was in the group of weak and medium EPL producers. The suppression of EPL production by PE was weaker. The obtained results suggest that EO or PE of Origanum vulgare L. may be a suitable tool for reducing yeast virulence, thus contributing to more effective therapy and prophylaxis of candidiasis. Candida albicans je oportúnny patogén, často zodpovedný za vznik kandidózy u ľudí. Jej patogenita je podmienená viacerými faktormi, ktoré sa podieľajú na vývoji rezistencie voči konvenčným antimykotikám, čo zhoršuje prognózu liečby. V štúdii sme sa zamerali na testovanie antimykotického účinku rastlinných extraktov (RE) a rastlinných silíc (RS) z čeľade Lamiaceae (Mentha piperita L., Thymus vulgaris L., Origanum vulgare L., Salvia officinalis L.). Následne sme sledovali vplyv antimykoticky najúčinnejšej silice a extraktu na produkciu extracelulárnej fosfolipázy (EPL) klinickými izolátmi Candida albicans. Z testovaných silíc bol lepší antimykotický účinok pozorovaný pri RS z Origanum vulgare L., Mentha piperita L. a Thymus vulgaris L., s priemernými hodnotami MIC 0,4 mg.ml-1. Z testovaných RE najlepší antimykotický efekt sme zaznamenali pri extrakte z Origanum vulgare L. s priemernou minimálnou inhibičnou koncentráciou (MIC) 4,9 mg.ml-1. Z 23 izolátov (92 %) tvoriacich EPL bola u 22 testovaná inhibícia produkcie EPL vplyvom RS a RE z Origanum vulgare L. Po expozícii buniek extraktom, 7 izolátov (31,8 %) produkovalo EPL intenzívne, 6 izolátov (27,3 %) vykazovalo strednú produkciu EPL a 8 izolátov (36,4 %) miernu. Jeden izolát (4,5 %) produkciu EPL nevykazoval. Po hodinovej expozícii buniek RS z Origanum vulgare L. (0,8 mg.ml-1) len 1 izolát (4,5 %) naďalej produkoval EPL intenzívne a až 15 izolátov (68,2 %) fosfolipázu netvorilo. Ďalšie 4 izoláty (18,3 %) produkovali EPL mierne a po jednom izoláte bolo v skupine slabých a stredných producentov EPL. Potlačenie produkcie EPL rastlinným extraktom bolo slabšie. Dosiahnuté výsledky naznačujú, že RS, prípadne RE z Origanum vulgare L. môže byť vhodným nástrojom na znižovanie virulencie kvasinky, čo môže prispievať k efektívnejšej terapii a profylaxii kandidóz.
