  2. 五種精油之成分分析及對馬拉色菌之抑制與抗發炎活性之研究 ; Study on the compositions, anti-Malassezia furfur and anti-inflammatory activities of five essential oils ; 本研究探討五種精油,分別是檸檬香茅(Cymbopogon citratus)、百里香(Thymus vulgaris)、甜馬鬱蘭(Origanum majorana)、突尼西亞迷迭香(Rosmarinus officinalis)及西班牙迷迭香(Rosmarinus officinalis)之抑制馬拉色菌能力、抗菌交互作用、抗發炎及抑制脂解?的活性評估,並以氣相層析質譜儀(GC/MS)分析五種精油的成分。 成分鑑定結果顯示,檸檬香茅精油主成分為geranial (41.66%)及neral (33.37%);百里香精油主成分為thymol (43.56%)、cymene (32.29%);甜馬鬱蘭精油主成分為terpinen-4-ol (41.01%)、cymene (11.02%);突尼西亞迷迭香精油主成分為1,8-cineole (54.57%)、camphor (8.58%)、α-pinene (8.42%);西班牙迷迭香精油主成分為1,8-cineole (38.26%)、α-pinene (22.41%)、camphor (10.45%)。 抗菌活性部分,檸檬香茅精油抗馬拉色菌( Malassezia furfur )效果最好,MIC90為353.2 ?g/mL;抗發炎試驗部分顯示,NO自由基清除能力中,檸檬香茅精油效果最好,SC50濃度為0.33mg/mL,抑制5-LOX活性中,甜馬鬱蘭效果最好IC50為36.73?g/mL;脂解?(Lipase) 抑制活性能力中,甜馬鬱蘭精油效果最好,其IC50為46.60?g/mL。 抗菌交互作用實驗中,檸檬香茅與甜馬鬱蘭組合、突尼西亞迷迭香與檸檬香茅組合、突尼西亞迷迭香與百里香組合以及西班牙迷迭香與百里香組合為無關性作用 (indifference),其餘組合皆為累加作用(additive)。 ; This study investigated the anti-M. furfur properties, synergism effect, anti-inflammatory and lipase inhibitory activities of five essential oils, lemongrass (Cymbopogon citratus), thyme (Thymus vulgaris), sweet marjoram (Origanum majorana), Tunisian rosemary (Rosmarinus officinalis) and Spanish rosemary (Rosmarinus officinalis).The compositions of five essential oils were analyzed by gas chromatography mass spectrometry (GC/MS). The results showed that the main components of lemongrass was geranial (41.66%) and neral (33.37%); thyme was thymol (43.56%)、cymene (32.29%); sweet marjoram was terpinen-4-ol (41.01%)、cymene (11.02%); Tunisian rosemary was 1,8-cineole (54.57%)、camphor (8.58%)、α-pinene (8.42%); and the Spanish rosemary was 1,8-cineole (38.26%)、α-pinene (22.41%)、camphor (10.45%). The lemongrass essential oil had the best anti-Malassezia furfur effect (MIC90 was 353.2 ?g/mL); and anti-inflammatory effect on NO free radical scavenging ability (SC50 was 0.33mg/mL).The sweet marjoram essential oil had the best effect on 5-LOX inhibitory activity (IC50 was 36.73?g/mL) and the lipase inhibitory activity (IC50 was 46.60?g/mL). In the synergism effect test, the mixture of lemongrass and sweet marjoram, Tunisian rosemary and lemongrass, Tunisian rosemary and thyme, and Spanish rosemary and thyme were indifference, the other combinations were additive effect.

  3. 綠花白千層之成分分析及對痤瘡桿菌之抑制與抗發炎活性之研究 ; Study on the compositions, anti-Cutibacterium acne and anti-inflammatory activites of Melaleuca quinquenervia essential oils ; 痤瘡為最常見的皮膚疾病之一,好發於面部、背部、手臂、胸部等等皮脂腺發達的部位,為了找出能有效控制痤瘡的物質,本實驗利用綠花白千層(Melaleuca quinquenervia) 的葉子,使用水蒸氣蒸餾法(SD)進行精油萃取,並進行成分分析及測定生物活性。利用氣相層析質譜分析儀(GC-MS)測定出萃取之綠花白千層精油之主成分為1,8-cineole 38.43%(醚類)、Azulene 17.21%(多環芳香烴)、D-Limonene 8.16%(單?類)、α-Pinene 7.70%(單?類);市售綠花白千層精油主成分為1,8-cineole 51.55%(醚類)、Azulene 8.16%(多環芳香烴) 、α-Terpineol 7.70%(單?烯醇)、D-Limonene 6.69% (單?類)、α-Pinene 6.41%(單?類),之後用這兩種精油測定其抗菌、抗發炎、脂肪?酵素抑制等活性分析,最後與抗生素進行協同作用的活性試驗,期望運用在化妝品上,增加其經濟價值。 抗菌活性試驗結果顯示,綠花白千層精油對於痤瘡桿菌(Cutibacterium acne)是有顯著抑菌的效果,5-LOX脂氧合?抑制能力與NO自由基清除的測試顯示,在特定濃度下,綠花白千層精油具備抗發炎的能力,脂肪?酵素抑制試驗結果顯示,綠花白千層精油能有效抑制脂肪?酵素活性,最後在精油與抗生素的協同作用實驗中,顯示綠花白千層與抗生素是有協同作用抗菌能力。 ; Acne is one of the most common skin diseases. It occurs on the face, back, arms, chest and other areas with active sebaceous glands. In order to find an effective material to control acne, this experiment used steam distillation(SD) extraction method to extract Melaleuca quinquenervia essential oils and then analyze it composition and biological activity. The results showed that the main components of the extracted Melaleuca quinquenervia were 1,8-cineole 38.43% (ether), Azulene 17.21% (Polycyclic Aromatic Hydrocarbons), D-Limonene 8.16% (monoterpene), and α-Pinene 7.70%(monoterpene). The main components of commercially Melaleuca quinquenervia essential oil are 1,8-cineole 51.55% (ether) ,Azulene 8.16%(polycyclic aromatic hydrocarbons), α-Terpineol 7.70% (monoterpenol), D-Limonene 6.69% (monoterpene) , and α-Pinene 6.41%(monoterpene) by gas chromatography-mass spectrometry (GC-MS).Next, these two essential oils are determine their antibacterial, 5- lipoxygenase inhibition ability , NO free radical scavenging and lipase inhibition , we also conduct the synergistic activity test antibiotics, and time-kill curve assay of essential oils and antibiotic. The results of the antibacterial activity test showed that Melaleuca quinquenervia essential oil has a significant antibacterial effect on Cutibacterium acne. The inhibition of 5-lipoxygenase and NO free radical scavenging ability showed that Melaleuca quinquenervia essential oil has anti-inflammatory effects. Melaleuca quinquenervia essential oil can effectively inhibit the activity of lipase and have synergistic effect with antibiotic.
